The lipids present in the plasma membrane of platelets are important for their function. Any changes in the lipid composition can affect the membrane receptor’s accessibility by drugs. The data on the lipid composition of platelet plasma membranes needed to be brought up to date and the impact of antiplatelet agents on plasma membrane composition was not well understood.
To address this, researchers isolated the platelet plasma membrane and analyzed its lipid composition using lipidomics. They studied the effect of ticagrelor, a P2Y12 antagonist, and its active metabolite on the lipid composition of platelet plasma membranes.
It was found that ticagrelor and its active metabolite increased cholesterol and phosphatidylcholine ether with short saturated acyl chains, and decreased phosphatidylcholine, indicating rigidification of the membrane.
